A psychological disorder is describe as any disturbance of emotional equilibrium, as manifested in maladaptive behavior and impaired functioning, caused by genetic, physical, chemical, biological, psychological, or social and cultural factors. While treating my client Bobby Lopez for about three months he has shown little signs of improvement, He started out with another therapist and was reassigned to me for a further study. Bobby stated that he started feeling down and didn’t really want to come out his house at times he would push his wife of three years away just because his mood was unstable, Bobby recently had a child who just turned 3 months he has had no interaction with playing or taking care of his child due to his mood. Before Bobby came to see a therapist he was drinking very heavily and would drink for sunrise to sundown to bypass the time and to make himself feel better. As we have been working on his drinking in order to help him to improve his mood disorder. The whole building is designed for energy efficiency, sustainability and environment friendly. The technology used to build the building will not only conserve energy but also increase its effectiveness. The semi-transparent screen used on the outside of the building, helps keep the interiors cool in summer and warm in winter. Answer: Smith Waits Characters: Smith, a young man of 20s Jane, an old woman of 70s The Unknown Man A Young Girl Act 1 Scene 1 The stage is bare and there are only few benches, all are empty. In front of those benches, there is a signboard, written Waiting Room. There is a man of around 20, who is sitting in a bench and is constantly having a look at the watch. He is wearing a trouser that is torn out and the shirts too, had few rusty stains, and at places, it too had torn out. There is a gate on the R, Jane, a woman of around 70, enters through RS, with a handbag and with a book in her hand. She started looking for a place to sit, when the young man makes a place for her. Jane: Thank you young man, I could not stand any more. Thank you for your help. What is your name? Young Man: My name is Smith Jones, maam. Jane: Oh! Good to meet you Smith, I mean Mr. Jones. Smith is sitting in the front bench and smoking. Jane is talking with a young girl. The girl is tired and so Jane had made place for her so that she could lie down and sleep. The young girl lay down, and soon she started snoring. Jane slowly moved to the front stage. Smith: So, you had your talk? Jane: (Smiling) A very lovely talk I must say. You know she had brown eyes. Smith: So? Jane: My mother too had brown eyes. I used to adore those brown eyes. Smith: Your eyes too are very beautiful and quite adorable. Jane: (laughing) I too was quite adorable in my young age. She too was waiting for someone; the person has thankfully arrived early. Smith: Who was your mother? Jane: Oh, she worked as a secret agent during the Second World War. (Whispering) Well, I will not tell you for, which country she worked. (The man, who came started becoming restless). Smith: (Angrily) Oh come on move now. You had your call, off with you now. Fermentation produces ATP with organic compounds like carbohydrates as the electron acceptor instead of oxygen, although less than cellular respiration. Yeast and muscle cells are capable of both cellular respiration and fermentation to harvest energy. When yeast cells are in a non-oxygen placement, the cells are forced to ferment. This is partially what it means when brewers ferment there beer. Our bodies use cellular respiration, to produce ATP, as our primary source of energy during normal activity. Although when we exert large amounts of energy all at once like running sprints would do, the normal cellular respiration would be unable to keep up with the required amount of oxygen to create ATP, therefore fermentation begins to assist making ATP. Cells use enzymes as biological catalysts to increase or accelerate the rate of reactions, such as those in photosynthesis or glycolysis. This allows reactions to occur under conditions that sustain life: An enzyme is simply a catalyst; also something extra that’s only purpose is to speed a process up that would otherwise take a very long time. Our bodies have copious amounts of these enzymes whose only purpose is to speed up needed reactions and bonds between separate chemicals throughout the body. This bonding process is completed through what is known as enzyme substrate interactions. (Enzyme, 2005) An enzyme-substrate is most easily explained as being an extremely specific key whole in which only a specific enzyme has access to, allowing that enzyme to enter the key whole and pushing to molecules into one. An enzyme-substrate complex substrate undergoes a chemical reaction forming a new product. The substrate still has the ability to break away from the enzyme at this point allowing it the ability to form with a different substrate. An enzyme product is simply a solution. The eventual mating of two chemicals to bond together forms a new enzyme product. Enzyme activity is regulated a couple of different ways, one is known as allosteric inhibition and the other is known as competitive inhibition. Competitive inhibitors bind with the active site forcing the substrate to compete with it, hence the title competitive inhibition. The companies which performed better in share market even in the crisis situations are generally perceived as better companies and the investors may invest more heavily in those shares. The share prices of better performing companies may not dip drastically even in unfavorable market conditions. In short, share price of a company is directly proportional to the growth of the company; when the company grows, the share prices will increase and when the company underperforms, share prices will decrease. Since dividend is a portion of annual profit made by a company, no company would be able to pay dividends to the shareholders when it underperforms. At the same time, it is not necessary that a company which is able to pay higher dividends is better than some other companies which are paying fewer dividends or no dividends at all. Ishmael says nothing about the causes of the war, or what each side was fighting for, or of the overall political and social conditions in Sierra Leone that caused the war. This was a deliberate strategy on the part of Beah, the author. He wanted to present the war through the eyes of a child. As a boy of twelve, when the war first affected him, he had no interest in politics. He had no reason to be interested—his main interest, understandably for a boy of his age, was in singing and dancing to rap music and hanging out with his friends. When the war comes to him, it is for him a battle for personal survival, not a political cause. He is also fueled by feelings of revenge—instilled into him by his army officers—against the rebels because they killed his family. Once again, these are personal feelings not political beliefs. For the reader, then, transported to a land he or she knows nothing about (for the American reader, that is), the war seems not only almost unimaginably brutal but also meaningless. It consists of one side mindlessly killing the other, and vice versa, in skirmishes in small villages. Ishmael does report Lieutenant Jabati’s speeches to his men, in which he says they are defending their country (â€Å"We kill them [the rebels] for the good and betterment of this country† [p. 123]), but such appeals to patriotism are not what inspire Ishmael. During the 1980s Sierra Leone was a one-party state governed by the All-People’s Congress (APC) party. However, this period was marked by extensive government corruption and abuse of power. Although Sierra Leone is rich in natural resources it became one of the poorest countries in the world because of mismanagement. The civil war in neighboring Liberia helped to create conditions for war in Sierra Leone because a Liberian war leader reportedly sponsored the rebel group called the Revolutionary United Front (RUF) as a way of destabilizing Sierra Leone, which at the time was a base for a United Nations peacekeeping force. The war broke out in 1991 in villages in eastern Sierra Leone that were near the Liberian border. The aim of the RUF was to seize and control the diamond sector, and in 1991 it took control of the diamond mines in the Kono district. (It is the mining area around Ishmael’s home town of Mogbwemo that the rebels seize in 1993.) In 1992 a military coup took place that established the National Provisional Ruling Council, replacing the civilian government. However, the new military government was powerless to prevent the RUF from controlling much of the country. It was the years immediately after this, from 1993 to January 1996, that Ishmael was a soldier. But this did not last, and the war dragged on, finally ending in January 2002, with the civilian government in charge. Some died before they could be tried, but in June 2007, the Special Court found three men guilty of war crimes, including not only murder, terrorism, and enslavement but also the act of conscripting or enlisting children under fifteen into the armed forces. As refugees from the war are slowly returning from neighboring countries, the Sierra Leone government is trying to create jobs and end political corruption. Revenues from diamond mining have increased significantly since the end of the war. Diamonds account for about half of Sierra Leone’s exports. However, Sierra Leone, with a population estimated in 2009 as 5,132,138, remains an extremely poor country with wide disparities in how wealth is distributed. According to the World Factbook, â€Å"The fate of the economy depends upon the maintenance of domestic peace and the continued receipt of substantial aid from abroad.† 4.How widespread is the use of child soldiers? It would be comforting to think that the forced conscription of children into the armed forces during the war in Sierra Leone was an aberration, not something that can happen again in the modern world. However, that is not the case. Even in the twenty-first century, the use of child soldiers is common in armed conflicts around the world. According to Human Rights watch, an international nongovernmental organization, as of 2007, there were an estimated 200,000 to 300,000 children fighting in various wars. According to a Global Report published in 2008 by the Coalition to Stop the Use of Child Soldiers, at the end of 2007 children were used as soldiers in seventeen armed conflicts around the globe. The coalition noted that this was down from twenty-seven conflicts in 2004, but the downturn was more because the conflicts had ended than because child soldiers were no longer being recruited. The Global Report identified the following countries where children were recruited for paramilitaries, militias, civilian defense forces or armed groups linked to or supported by governments: Chad, Colombia, Democratic Republic of the Congo, India, Iran, Ivory Coast, Libya, Myanmar, Peru, Philippines, Sri Lanka, Sudan, and Uganda. The most flagrant offender, according to the Global Report, is Myanmar, where the government uses thousands of children in its battle against rebel groups. In Uganda, tens of thousands of children have been forced into joining armies over a period of nearly twenty-five years. In some of these countries, including Uganda, girls as well as boys have been forced to become soldiers. There have in recent years been concerted international efforts to end the use of child soldiers. Sierra Leone, which has tried and convicted men responsible for recruiting child soldiers, has become a leader in this issue. The use of child soldiers has now been prohibited by international law. The Optional Protocol to the Convention on the Rights of the Child on the involvement of children in armed conflict has been ratified by 120 states. The UN Security Council has adopted resolutions calling for the establishment of a monitoring mechanism on children and armed conflict. However, these and other prohibitions are no guarantee that when new conflicts break out, child soldiers will not be recruited. 5.Is Beah’s story factually accurate? A Long Way Gone achieved popular and critical success, but questions have been raised by some regarding the factual accuracy of a number of events Beah recounts in the book. Beah writes that his village was attacked in January 1993 and after that he became a refugee from the war. Critics claim that there are school records showing that Beah was in school later than this date, and that the village was attacked in 1995, not 1993. This would mean that Beah would actually have been recruited at the age of fifteen, not thirteen as he writes in the book. This would have meant that he was only a child soldier for a few months, rather than over two years. Some critics point to the structure of the book to confirm this. They point out that most of the book deals with Beah’s wanderings as a refugee and the months he spent in rehabilitation. Only two chapters (13 and 14) cover his actual experiences as a soldier (although he does present more incidents from his military service at various points in flashbacks). Questions have also been raised about the account Beah gives of the fight between the former boy soldiers at the rehabilitation home, in which several boys were killed. In 1879 Tomas Edison invented the electric light, I guess you could say he just lit the way for may other inventions that people made during this time period. The progressives where middle class people that where mostly composed of young people who wanted things to change, they believed that educating people was the best way to overcome a problem. They formed volunteer organizations that people would come to and they would educate them on what they thought the problem was with something. Some of the organizations I found where: (American Bar Association, U.S Chamber of Commerce, National Association for the advancement of Colored People, and many others. I think that they did this to increase the awareness of a problem and try to fight that problem with strength in numbers and educations, both are very strong weapons when tackling a problem. Muckraking was another way people got their point across, they would find out all the information that they could on a person and they would basically, just drag their good name thought the mud, telling the people about, the things that they might not know, the corrupt things and bad deals and kickbacks...

Pip is the main character of this story. The whole plot of the novel revolves around him and his metamorphosis into an adult from an immature adolescent.  1. Immature: pip was highly immature. This characteristic in him can be clearly seen in pg 59 where he says † but when she was gone, I looked about me for a place to hide my face in and got behind one of the gates in the brewery lane and leaned my sleeve angst the wall there, and leaned my forehead on it and cried. As I cried, I kicked the wall and took a hard twist at my hair; so bitter were my feelings so sharp were the smart without a name that needed counteraction.† Pip cries so much, suddenly begins to have such a low self esteem, why? Just because a girl came and commented that he had coarse hands, wore thick boots and â€Å"called the knaves Jack.† Instead of standing up for himself, pip simply cries. This goes to show how immature pip was.  Pip’s immaturity is also seen in his â€Å"romantic idealism.† Ever since his first meeting with beautiful Estella, he always thought of her in page 101 he says, â€Å"often after dark, when I was pulling the bellows for Joe, and we were singing Old Clem, and the thought how we used to sing at Miss Haveshams would seem to show me Estella’s face in the fire, with her pretty hair fluttering in the wind and her face scorning at me. Often at such a time I would look towards those panels of black night in the wall which the wooden windows then were, and would fancy that I saw her drawing her face away and would believe that she had come at last.† In spite of Estella being s o insulting he instantly falls for her, rather for her looks. This again shows the immaturity in pip. 2. Generous and sympathetic: pip was generous and sympathetic, a fact that can be seen in his numerous acts of kindness throughout the novel. In page ___ he sympathizes with Magwitch and helps him escape. A small boy going out of his way to help a wretched convict! This goes to show how sympathetic pip was.  Not just sympathetic, pip was large at heart. He was generous. This can be witnessed in page _____ where he secretly buys Herbert’s way into business. The third example to show that pip was generous is in page 137where he says, â€Å"as I passed the church I felt a sublime compassion for the poor creatures who were destined to go there, Sunday after Sunday, all their lives thru and to lie obscurely at last among the low green mounds. I promised myself that I would do something for them one of these days and formed a plan in outline for bestowing a dinner of roast beef and plum pudding, a pint of ale and a gallon of condescension upon everybody in the village.† 3. Had a great desire for self-improvement: pip always wanted to better himself, better his condition and better his status. This trait in him can be witnessed in page 66 where he tells Joe, â€Å"and that there had been a beautiful young lady at miss Havesham’s who was dreadfully proud and that she had said I was common, and that I wished I was not common, and that the lies had come of it somehow, though I didn’t know how.† Moreover his deep desire to improve in life, to be â€Å"Uncommon† is also seen in page 68 where he goes to one of his relatives, Biddy to get educated from her. He says, â€Å"when I woke up the best step I could take towards making myself uncommon was to get out of Biddy everything she knew.†Ã‚  In page 164 he asks Herbert to correct his mannerisms if he found them un-gentlemanly. He says, â€Å"I further mentioned that as I had been brought up a blacksmith in a country place, and knew very little of the ways of politeness, I would take it is a great kindness in him i.e. Herbert if he would give me a hint when ever he saw me going at a loss or going wrong.†

GFP Transformation Into E Coli Biology Essay Genetic transformation  is the technique of introducing a recombinant DNA into a living cell. In this experiment, we introduced pGLO plasmid into E. Coli bacteria through the heat-shock method. CaCl2 solution was used to make the E. coli cells competent. Intruduction Genetic transformation is the technique involving introduction and expression of foreign(exogenous) DNA in a living host cells. Scientists cut out interested gene from human, plants or animals DNA, insert it into a vector to produce a recombinant DNA and introduce the recombinant DNA into host cells which express the exogenous genes and produce interested proteins under appropriate condition. Demonstrated by Frederick Griffith in 1928, transformation has been applied in various areas of biotechonology. For example, genes coding for human insulin can be genetically transformed into bacterial cells. In this way, genetic transformation allows the production of protein products on a large scale. The introduction of a foreign DNA into a host cell requires the use of a vector. Vectors are small DNA molecules that can be used to combined with foreign genes and transfer them into the host cells. In a laboratory research, plasmid is one of the most commonly used vectors to transform foreign DNA. The recombinant plasmid used in this experiment is pGLO plasmids. pGLO plasmids contain  two genes that are useful: the gene coding for beta-lactamase and the gene coding for the green  ¬Ã¢â‚¬Å¡uorescent protein(GFP). The expression of beta-lactamase gene provides resistance to the antibiotic ampicillin. GFP is extracted from Aequorea victoria (bioluminescent jelly fish). It is a polypeptide consisting of 238 amino acid(Cubitt et al., 1995). For its non-invasive characteristic and capacity of resisting interferences, GFP has been widely used as a visual marker for gene expression for more than 20 years(Gilbert et al., 2000). Even under normal light, GFP presents yellow-green color that can be easily observed. The objects of this experiment were to perform a transformation of E. coli with a plasmid containing the GFP DNA gene and force the GPF to be expressed in a particular environment. In this experiment, we induced E. coli to take in pGLO into their cytoplasm and grew the E. coli cells in different plates. CaCl2 solution is utilized as transformation solution to increase cell membrane  permeability, thus plasmid vectors are able to enter the cell. The ampicillin-resistance gene in the plasmid was utilized as the selectable marker, which means E. coli cells transformed with plasmid are able to grow in an environment with ampicilin. Because the GPF expression is driven by the arabinose-stimulated PBAD promoter, it is expected that the E. coli cells grown in the plate with sugar arabinose would express GPF DNA and present a yellow-green color in the UV light. Materials and Methods Plasmid Transformation Plasmid could be easily taken in by E. coli cells if the cells had been treated with calcium salt. First of all, two micro test tubes were labeled with either +pGLO or -pGLO. Each tube was added with 250ÃŽ ¼L of CaCl2 solution and then placed on ice. A single colony of E. coli bacteria was added into each tube. The cells were dissolved into the solution by gently spinning the tubes. Then both tubes were placed on ice again. After the entire colony was dispersed, the tubes were examined with UV light to make sure that there were no visible green clumps of cells in the solution. Then 10ÃŽ ¼L of pGLO plasmid was added into the +pGLO tube and gently mixed, while no plasmid was added into -pGLO tube. Both tubes were placed on ice for 10 minutes. Following the 10-min incubation, the tubes were heat shocked. Both tubes were transferred into 42 °C water bath for exactly 50 seconds, and then immediately placed back on ice for 10 minutes. The heat shock process must be rapid. After heat shock, 250ÃŽ ¼L of Luria broth (LB) were added to each tube and the tubes were placed at room temperature for 10 minutes. Selection of GFP Transformation To grow and select the cells with GFP DNA, four agar plates were obtained: 1 LB, 2 LB/AMP and 1 LB/AMP/arabinose. Among them, 1 LB/AMP plate and 1 LB/AMP/arabinose plate were labeled with +pGLO, while 1 LB plate and 1 LB/AMP plate were labeled with -pGLO. 100ÃŽ ¼L of cells from +pGLO tube was added to each of the plates labeled with +pGLO, while 100ÃŽ ¼L of cells from -pGLO was added to each of the plates labeled with -pGLO. New sterile loops was used for each plate. The whole process was conducted near fire. Then four plates were stached upside down at 37 °C for 24 hours. A photo of the plates were taken in the UV light the next day. Results Figure 1 on the next page shows a photo of four plates. Table 1on the next page illustrates the observation results of each plate. From the photo, we can observe the growth condition of colonies on each of the four plate. Plate 1: the untransformed E. coli colonies grew normally and form a lawn on the plate; Plate 2: no colonies grew; Plate 3: transformed E. coli colonies grew and fluorescenced yellow-green in UV light; Plate 4: E. coli colonies grew and presented white color in UV light. Fig Photo was taken after the plates had been incubate at 37 °C for 24 hours. The cells grew normally as in natural condition. Plate 1 is a negative control which excludes possible contaminants. In the plate containing LB/amp and E.coli -pGLO(Plate 2), no colonies grew, because the ampicillin  in the medium killed the cells by inhibiting the cell wall from producing. In the plate containing LB/amp/ara and E.coli +pGLO(Plate 3), colonies were grew, because these bacterial cells contains plasmid carrying ampR gene which perform a resistance against ampicillin. Under UV light, the colonies emit green-yellow fluorescence, because the GFP gene on plasmid was expressed in an environment with arabinose. In the plate containing LB/amp and E.coli +pGLO(Plate 4), white colonies were grew because of the presence of ampR gene and absence of arabinose. The presence of ampicillin in the medium is to identify if the E.coli cells have taken in plasmid and thus acquired ampicillin-resistance gene on the plasmid. The results of Plate 4 compared with Plate 2 indicate that E.coli cells have taken in plasmid successfully. The presence of arabinose is to identify if the E.coli cells containing recombinant plasmid successfully inserted with GFP gene. The results of Plate 3 compared with Plate 4 indicate that the plasmid was successfully recombined, thus the cells express GFP under the induction of arabinose. Conclusion In this experiment, GFP transformation in E. coli was perform in order to examine how the recombinant plasmid can be introduced into bacterial cells, incorporated into bacterial genome and express recombinant protein. The experiment shows the expected results, which successfully support the hypothesis.

Place You Have Visited for a Holiday

Cameron Highlands is the smallest district in the state of Pahang which is located in the north-western corner of the state. One of the wonders of Malaysia, it is the largest and most famous hill resort in the country. This highland paradise still retains much of the charm of an English village. Being a primarily agricultural domain, you will find an abundance of vegetables and fruits farms here. Cameron Highlands is also the leading producer of flowers and tea in Malaysia. Be prepared for a lovely sight of extraordinary flowers you won’t see flourishing elsewhere in Malaysia!Located on the Titiwangsa Range at about 1500 metres above sea-level, the temperature here can drop to 16 ? C or lower at certain times. Cameron Highlands is made up of 4 main townships followed by smaller settlements at different elevations. The first town from the south is Ringlet, followed by Tanah Rata, Brinchang, Kea Farm, Tringkap, Kuala Terla and Kampung Raja. Ringlet If you're driving via the old road from Tapah, this is the first town you will reach. Located at an elevation of 1,200 metres above sea-level, Ringlet is one of the main agricultural hub for the highlands.Bertam Valley  which is located about 5km away is one of the main flower and vegetable producers in Cameron highlands. You wont find any hotels here in Ringlet. The nearest hotel is the LakeHouse which is less then 10 minutes away. This is also the place to fill up on fuel if you're low on supply. Tanah Rata Further up is Tanah Rata which is about 12 km’s (20 minutes drive) from Ringlet. This is the administrative capital of Cameron Highlands where the government offices, hospital and the district police station is located.The main bus station is also located here and this is where you would be able to get your transportation needs within and out of the highlands sorted out. Most of the chalets and backpacker's accommodation is located here in and around Tanah Rata, Almost all the jungle trekking and w aterfall trails starts from here. Local tours and transfers out of Cameron Highlands can be obtained from the various tour operators in town. Money changers and internet cafe's can also be found here. Tanah Rata would be a nice place to enjoy the highlands, especially during the weekdays.It is much more quieter compared to Brinchang where most of the big hotels and restaurants are located. Brinchang Next is Brinchang which is about 4 km’s (10 minutes drive) from Tanah Rata. This is where you will find most of the hotels and restaurants. The night market which is open every Saturday night and on holidays, features all kinds of vegetables, flowers, fruits and other products of the highlands. This place can get really crowded during the weekends and holiday season. Traffic jams and parking problems can be a nightmare during this time.Next to Ringlet, this is the only other town with petrol stations so keep your fuel tank in check. Kea Farm, Tringkap, Kuala Terla & Kampung Raja B eyond Brinchang, the road leads to all the way to Blue Valley. Most of the tourist spots are located around this route. This is where you will find the markets, vegetable farms, flower gardens, bee gardens and butterfly gardens. The road also branches out to the Sungai Palas tea plantation from here where you can visit the factory or have a cup of tea while enjoying the breathtaking view of the plantation.If you continue on the main road heading north, it will be downhill from Kea Farm onwards. You will see vegetables farms all the way from here to Tringkap and beyond. Tringkap is a small farming community where you can find stalls along the road selling local produce. Fifteen minutes away is Kuala Terla, another small farming community. Kampung Raja is the last town on the map. Since the opening of the Simpang Pulai road, this town has become the gateway to the highlands from the north. From here, you can head west to Ipoh or east to Gua Musang in Kelantan.
